ABSTRACT

Objective To clone express and purify Schistosoma japonicum fructose?1 6?bisphosphate aldolase SjFBPA in E. coli and observe its expression in different developmental stages of S. japonicum. Methods FBPA gene was amplified from S. japonicum adult worm cDNA by using PCR. The amplified product was recombined into pET28a plasmid and inducibly expressed with IPTG in E. coli BL21. SDS?PAGE and Western blotting were employed to analyze and identify the recombinant protein SjFBPA rSjFBPA . Then rSjFBPA was purified by chromatographic purification and its purity was analyzed by SDS?PAGE. The protein concentration of rSjFBPA purified was measured by the BCA method. Furthermore SjFBPA mRNA was ana?lyzed in different developmental stages of S. japonicum by RT?PCR. Results SjFBPA was successfully amplified by using PCR and identified by restriction enzyme digestion and sequencing. The Western blotting analysis confirmed that the recombinant pro?tein could specifically reactive to the anti?His?tag monoclonal antibody. The concentration of the purified recombinant protein was about 4 mg/ml. The result of RT?PCR showed that SjFBPA mRNA was expressed in cercaria schistosomulum adult worm and egg of S. japonicum. Conclusion SjFBPA is successfully recombined and expressed in a prokaryotic system and SjFBPA mRNA is expressed in cercaria schistosomulum adult worm and egg of S. japonicum.

ABSTRACT

<p><b>OBJECTIVE</b>To seek effective drugs inhibiting herpes simplex virus (HSV-2) with the signal pathway required by virus replication as the target spot.</p><p><b>METHOD</b>HSV-2-induced Vero cytopathic effect was observed, and MTT method was adopted to detect call activity, in order to assess the antiviral capacity of freeze dried powder of aqueous extracts of Saururus chinensis (AESC). Western blot was used to check the effect of AESC on signal pathway induced by HSV-2 virus in HeLa cells.</p><p><b>RESULT</b>AESC obviously inhibits the pathway activation of CPE induced by HSV-2 infection and NF-kappaB required for virus replication. The inhibition ratio of AESC freeze dried powder at 0.10, 0.03, 0.01 and 0.003 g x L(-1) were (70.68 +/- 3.39)%, (61.74 +/- 2.13)%, (39.31 +/- 1.10)% and (18.54 +/- 3.44)%, respectively. The IC50 was determined at (0.023 +/- 0.004) g x L(-1). The inhibition concentration of the positive control acyclovir was 0.001 g x L(-1) (5.0 x 10(-6) mol x L(-1)). The best administration time was from 2 h before infection to 6 h after infection. Western blot also showed that AESC can notably inhibit HSV-2-induced NF-kappaB nuclear transfer.</p><p><b>CONCLUSION</b>AESC can inhibit HSV-2 virus replication, which is related to the pathway activation of NF-kappaB required for virus replication.</p>

ABSTRACT

BACKGROUND: There is often a physical and psychosocial effect in burn injury, and the degree of mental stress reaction is related to social support, individual experience and personality characteristics.OBJECTIVE: To investigate the mental stress reaction, the degree of social support and the Minnesota Multiphasic Personality Inventory (MMPI)personality characteristics in acute burn patients, and to analysis the influence factor of mental stress reaction.DESIGN: Cross-sectional study.SETTING: West China Mental Health Center of Sichuan University, Burn Department of Mianyang Municipal Central Hospital, and Burn Department of General Hospital of Panzhihua Iron and Steel Company.PARTICIPANTS: From April 2003 to April 2005, totally 104 mild, moderate or severe burn patients, including 92 males and 12 females, aged 12-60 years, selected from Mianyang Central Hospital and Department of burn of General Hospital of Panzhihua Iron & Steel Co., Ltd took part in the study.METHODS: The emotional problems and personality characteristics were measured with the Hamilton Rating Scale for Depression (HAMD), the Hamilton Rating Scale for Anxiety (HAMA), the Minnesote Multiphasic Personality Inventory (MMPI), the Social Support Rating Scale (SS) as well as general clinical data,by the rating scales 3 day after burn. The patients were divided into anxiety-depression group (n=51) and normal emotion group (n=53) according to their anxiety-depression scores (anxious score> 7,depressive score> 8), those influencing factor included burning total area,age, sex and society support, Stepwise Regression was introduced to get the main factors that influenced mental stress reaction.MAIN OUTCOME MEASURES: ① Scores of each scale and comparison of general clinical data in each group; ② stepwise regression analysis of depression and anxiety of burn patients.RESULTS: Data of Totally 104 patients was entered the final analysis. ①The scores were 10.4±2.46 and 16.8±5.94 of anxious and depressive patients during acute burn period, which were higher than those in normal group (4.29±2.26, 5.48±2.06, P=0.01). ② The main influential factor for depression were burning area and social supporting availability, and those for anxiety were age, burning area, social supporting availability and subjective social supporting degree. ③ MMPI test results show that T scores in anxiety-depression group were below 60 points, however T-scores were above 60 points in normal group including cheating points, hypomania,schizophrenia, psychasthenia and paranoid ideation CONCLUSION: Burn is a mental crisis to burn patients, which can lead to abnormal emotional reaction and personality change. The depressive and anxious score are positively related with burning area, and are negatively correlated with society supporting availability. The good social support system is benefit to over mental crisis period.

ABSTRACT

Purpose:To evaluate two different methods of radiotherapy, which affect the results of cervical cancers treated and complications of the rectum or bladder. Methods:From July 1991 to December 1996, 124 cases of cervical cancer were randomly allocated into two groups. Group A (59 cases:stage Ⅰ 1, Ⅱa 12, Ⅱb 30, Ⅲa 11, Ⅲb 5) received 45—55 Gy of external radiation(ER) to the parametrium and 50—65 Gy of intracavitary irradiation (4—5 Gy per fraction, 2 fractions per week). Group B(65 cases: stage Ⅰ 2, Ⅱa 15, Ⅱb 29, Ⅲa 10, Ⅲb 9) received 26—40 Gy ER to the whole pelvic and 30—40 Gy of intracavitary irradiation (4—5 Gy per fraction, 2 fractions per week), an additional 15—29 Gy ER to the parametrium. Results:In group A, 3 year and 5 year survival rates were 81.4% and 71.7%. In group B, they were 84.6% and 70.8%. The rates of complication for the rectum were 25.4%(3 year) and 21.7%(5 year) in group A, 10.8%(3 year) and 8.3%(5 year) in group B. The rates of rectal complication( P
